There are about 30 filled soul gems available for the taking in the College of Winterhold (check both the Arch-Mage's Quarters and all the dorm rooms). Once you take them all, simply wait two weeks (takes about ten minutes real time) and then all of the gems will have respawned. For me, this has been the easiest way to get soul gems yet.
Azuras star is refillable after you use it. I don’t remember which will fill first but it’s not a major loss if you test it vs a petty soul gem. Azura's star will be filled last. You can find two filled Grand soul gems in the lobby of the Arcane University, and two greater soul filled gems in the Chorrol Mages guild (in grand gems).For grand souls the easiest creatures are Mammoths (but not Giants they give greater souls). The rest of the Grand group are high level falmer or draugr and dragon priests which are all harder to find consistently. For enchanting you can skip the soul trap all together. Soul can't fill gem of lower size. Grand soul will newer fill petty gem, but petty soul can fill grand gem. Make sure that won't happen and you have appropriate size gems in your inventory.
